A preliminary analysis of death cause, capture-related mortality, and survival of adult red deer in northwestern Patagonia Ecología austral
Flueck,Werner T; Smith-Flueck,Joanne M; Bonino,Never A.
The red deer (Cervus elaphus), among the world´s 14 most invasive exotic mammals, has recently arrived in Patagonia. Forty-seven deer were captured, marked with radio collars, and monitored in order to determine survival rates and identify causes of death. Net gunning from a helicopter allowed captures to be evenly distributed through the study area and in a timely manner. The absence of capture related mortality in our study agrees with previously reported low rates for this method. Animals were monitored for periods ranging from 5 to 2611 days. Ten animals provided censored values due to radio failures or being shot: they survived on average 811 days (SE = 221) after capture. Ten animals experienced natural deaths with an average time alive after capture...

Adhesion and invasion of Clostridium perfringens type A into epithelial cells BJM
Llanco,Luis A.; Nakano,Viviane; Moraes,Claudia T.P. de; Piazza,Roxane M.F.; Avila-Campos,Mario J..
ABSTRACT Clostridium perfringens is the causative agent for necrotic enteritis. It secretes the major virulence factors, and α- and NetB-toxins that are responsible for intestinal lesions. The TpeL toxin affects cell morphology by producing myonecrosis, but its role in the pathogenesis of necrotic enteritis is unclear. In this study, the presence of netB and tpeL genes in C. perfringens type A strains isolated from chickens with necrotic enteritis, their cytotoxic effects and role in adhesion and invasion of epithelial cells were evaluated. Six (27.3%) of the 22 C. perfringens type A strains were harboring the tpeL gene and produced morphological alterations in Vero cells after 6 h of incubation. Strains tpeL (-) induced strong cell rounding after 6 h of...

Adhesive and invasive capacities of Edwarsiella tarda isolated from South American sea lion BJM
Fernández,Araceli; Paz Villanueva,María; González,Mario; Fernández,Fabiola; Latif,Fadua; Flores,Sandra Nonier; Fernández,Heriberto.
Edwarsiella tarda is a zoonotic bacterium that can be isolated from humans, animals and the environment. Although E. tarda is primarily considered a fish pathogen, it is the only species of its genus considered to be pathogenic for humans as well. A survey of zoonotic intestinal bacteria in fresh feces from South American sea lions (SASL) Otaria flavescens, reported E. tarda as the most frequently isolated species. In this study, we used HEp-2 cells to establish in vitro the adherence and invasive ability of 17 E. tarda strains isolated from SASL fecal material. All the strains were able to adhere and invade HEp-2 cells with adhesion and invasion percentages ranging from 56 to 100% and 21 to 74%, respectively. Despite the expression of these pathogenic...

Adipose tissue-derived stem cells promote pancreatic cancer cell proliferation and invasion BJMBR
Ji,S.Q.; Cao,J.; Zhang,Q.Y.; Li,Y.Y.; Yan,Y.Q.; Yu,F.X..
To explore the effects of adipose tissue-derived stem cells (ADSCs) on the proliferation and invasion of pancreatic cancer cells in vitroand the possible mechanism involved, ADSCs were cocultured with pancreatic cancer cells, and a cell counting kit (CCK-8) was used to detect the proliferation of pancreatic cancer cells. ELISA was used to determine the concentration of stromal cell-derived factor-1 (SDF-1) in the supernatants. RT-PCR was performed to detect the expression of the chemokine receptor CXCR4 in pancreatic cancer cells and ADSCs. An in vitro invasion assay was used to measure invasion of pancreatic cancer cells. SDF-1 was detected in the supernatants of ADSCs, but not in pancreatic cancer cells. Higher CXCR4 mRNA levels were detected in the...

The cross-reactive antigens of Neospora caninum and Toxoplasma gondii are important in the exploration to determine the common mechanisms of parasite-host interaction. In this study, a gene encoding N. caninum apical membrane antigen 1 (NcAMA1) was identified by immunoscreening of a N. caninum tachyzoite cDNA expression library with antisera. from mice immunized with recombinant T gondii apical membrane antigen 1 (TgAMA 1). NcAMA1 was encoded by an open reading frame of 1695 bp, which encoded a protein of 564 amino acids. The single-copy NcAMA1 gene was interrupted by seven introns. NcAMA1 showed 73.6% amino acid identity to TgAMA1. Mouse polyclonal antibodies raised against the recombinant NcAMA1 (rNcAMA1) recognized a 69-kDa native parasite protein by...

BACKGROUND: Bacterial pathogens have many strategies for infecting and persisting in host cells. Adhesion, invasion and intracellular life are important features in the biology of mollicutes. The intracellular location ofUreaplasma diversum may trigger disturbances in the host cell. This includes activation or inhibition of pro and anti-apoptotic factors, which facilitate the development of host damage. The aim of the present study was to associate U. diversum infection in HEp-2 cells and apoptosis induction. Cells were infected for 72hs with four U. diversum clinical isolates and an ATCC strain. The U. diversuminvasion was analyzed by Confocal Laser Scanning Microscopy and gentamicin invasion assay. The apoptosis was evaluated using pro-apoptotic and...

Babesia bovis: Effects of cysteine protease inhibitors on in vitro growth OAK
Okubo, Kazuhiro; Yokoyama, Naoaki; Govind, Yadav; Alhassan, Andy; Igarashi, Ikuo.
In the present study, we examined the effects of four kinds of cysteine protease inhibitors (E64, E64d, leupeptin, and ALLN) on the in vitro asexual growth of Babesia bovis. Of these, only the lipophilic inhibitors, E64d and ALLN, were found to effectively inhibit the growth of B. bovis. In further experiments, E64d, but not ALLN, significantly suppressed the parasite’s invasion of host erythrocytes, while both chemicals, especially ALLN, inhibited the parasite’s replication within the infected erythrocytes. These data suggested the presence of cysteine protease(s) derived from B. bovis, in which the protease(s) would play important roles in the erythrocyte invasion and/or replication processes of the parasite.

Cantharidin suppressed breast cancer MDA-MB-231 cell growth and migration by inhibiting MAPK signaling pathway BJMBR
Gu,X.-D.; Xu,L.-L; Zhao,H.; Gu,J.-Z; Xie,X.-H.
As an active constituent of the beetle Mylabris used in traditional Chinese medicine, cantharidin is a potent and selective inhibitor of protein phosphatase 2A (PP2A) that plays a crucial role in cell cycle progression, apoptosis, and cell fate. The role and possible mechanisms exerted by cantharidin in cell growth and metastasis of breast cancer were investigated in this study. Cantharidin was found to inhibit cell viability and clonogenic potential in a time- and dose-dependent manner. Cell cycle analysis revealed that cell percentage in G2/M phase decreased, whereas cells in S and G1 phases progressively accumulated with the increasing doses of cantharidin treatment. In a xenograft model of breast cancer, cantharidin inhibited tumor growth in a...

Distribution of virulence genes sefC, pefA and spvC in Salmonella Enteritidis phage type 4 strains isolated in Brazil BJM
Castilla,Karina Salvagni; Ferreira,Claudete Serrano Astolfi; Moreno,Andrea Micke; Nunes,Iolanda Aparecida; Ferreira,Antônio José Piantino.
The distribution of virulence genes, sefC, pefA and spvC, was investigated in 110 Salmonella Enteritidis phage type 4 strains by polymerase chain reaction. Their influence in the caecal colonization and invasion of liver and spleen of one-day-old chickens was studied. Eight isolates were negative for the spvC gene, three for the pefA gene and one, for the sefC gene. These results allowed grouping the strains into four genotypes. Presence of these genes did not influence bacteria invasion in the liver and spleen of the chickens ten days after infection, although the presence of more than one fimbrial gene can be related to caecal colonization.

Downregulation of survivin by siRNA inhibits invasion and promotes apoptosis in neuroblastoma SH-SY5Y cells BJMBR
Zhang,L.; Liang,H.; Cao,W.; Xu,R.; Ju,X.L..
Neuroblastoma is a solid tumor that occurs mainly in children. Malignant neuroblastomas have a poor prognosis because conventional chemotherapeutic agents are not very effective. Survivin, a member of the inhibitor of the apoptosis protein family, plays a significant role in cell division, inhibition of apoptosis, and promotion of cell proliferation and invasion. Previous studies found that survivin is highly expressed in some malignant neuroblastomas and is correlated with poor prognosis. The aim of this study was to investigate whether survivin could serve as a potential therapeutic target of human neuroblastoma. We employed RNA interference to reduce survivin expression in the human neuroblastoma SH-SY5Y cell line and analyzed the effect of RNA...

Expression of CHPF modulates cell proliferation and invasion in lung cancer BJMBR
Cao,Chengsong; Liu,Yong; Wang,Qun; Zhao,Jing; Shi,Ming; Zheng,Junnian.
Lung cancer is the most common malignancy worldwide and is characterized by rapid progression, aggressive behavior, frequent recurrence, and poor prognosis. The TCGA database indicates that chondroitin polymerizing factor (CHPF) is overexpressed in human lung cancer tissues compared with normal tissues and this overexpression corresponds to shorter overall survival in lung cancer patients. In this study, to investigate the function of CHPF in lung cancer, lentiviral vectors expressing CHPF shRNA were stably transduced into A549 and H1299 cells. Compared to shCtrl cells, CHPF knockdown cells had significantly reduced proliferation. Furthermore, the silencing of CHPF in A549 and H1299 cells resulted in apoptotic induction, which led to decreased colony...

Foraging behavior interactions between the invasive Nile Tilapia (Cichliformes: Cichlidae) and three large native predators Neotropical Ichthyology
Birck,Tiago; Message,Hugo José; Baumgartner,Gilmar; Sebastien,Nyamien Yahaut; Baumgartner,Dirceu.
ABSTRACT The predator-prey relationships between juvenile Nile Tilapia Oreochromis niloticus and native fish species of the Paraná River basin, Brazil, were experimentally examined. Juveniles of O. niloticus were offered to three native predator species (Salminus brasiliensis, Pseudoplatystoma corruscans, and Brycon orbignyanus) in 2,000-L tanks with four levels of habitat complexity (0%, 50%, 100% and RD (rocks and driftwood)). Predator efficiency was more variable among species (S. brasiliensis consumed 86.6% of the prey, P. corruscans 22.5% and B. orbignyanus 18.3%) than among levels of habitat complexity, and S. brasiliensis was faster than the others in detecting and consuming the prey. The higher predatory efficiency observed for S. brasiliensis can...

Ichthyofauna of headwater streams from the rio Ribeira de Iguape basin, at the boundaries of the Ponta Grossa Arch, Paraná, Brazil Biota Neotropica
Frota,Augusto; Message,Hugo José; Oliveira,Rachel Calil de; Benedito,Evanilde; Graça,Weferson Júnio da.
Abstract: The rio Ribeira de Iguape basin (Paraná State) is highly relevant for the preservation of remnants of the Atlantic Forest. Its high levels of diversity and fish endemism explain the river dynamics promoted by the Ponta Grossa Arch, a geological structure that promotes headwater capture and isolation between the upper rio Paraná drainage, the rio Iguaçu and the rio Ribeira de Iguape. Here, our objective was to provide an unprecedented inventory for headwater streams of the rio Ribeira de Iguape basin at the boundaries of the Ponta Grossa Arch. We found 29 species of fish representing four orders and nine families. Siluriformes was the richest order followed by Characiformes. Nine species presented high abundance of sampled individuals,...

Interleukin-27 augments the inhibitory effects of sorafenib on bladder cancer cells BJMBR
Cao,J.Y.; Yin,H.S.; Li,H.S.; Yu,X.Q.; Han,X..
Both sorafenib and interleukin-27 (IL-27) are antineoplastic drugs. This study aimed to investigate the synergistic effect of these two drugs on bladder cancer cells. HTB-9 and T24 cells were stimulated with IL-27 (50 ng/mL), sorafenib (2 μM) or the synergistic action of these two drugs. The cells without treatment acted as control. Cell proliferation, apoptosis and invasion were measured by bromodeoxyuridine assay, flow cytometry and modified Boyden chamber, respectively. Simultaneously, both modified Boyden chamber and scratch assay were used to assess cell migration. Finally, the phosphorylation levels of key kinases in the Akt/mechanistic target of rapamycin (mTOR)/mitogen-activated protein kinase (MAPK) pathway, and expression levels of matrix...

Invasion of Matrix Species in Small Habitat Patches Ecology and Society
The diversity, expressed as species richness in equal-sized samples, of wood-living beetles in successional stages of deciduous forest after forest fires approximately 100 years ago did not differ between patch sizes in this study. Thus, the crucial question for conservation is whether the lack of reduction in diversity in small patches means that a number of small patches can elevate the regional diversity as efficiently as can one larger area. In the present study, the smaller patches did not differ from larger patches in substrate availability, quality, or heterogeneity. The frequency of a group of species was measured as the number of occurrences, viz. the number of trees on which a species was found, summed over all species in the group. The number of...

Invasiveness of Cut-Leaf Ground-Cherry (Physalis angulata L.) Populations and Impact of Soil Water and Nutrient Availability Chilean J. Agric. Res.
Travlos,Ilias S.
Biological invasions are a major threat to natural ecosystems and agroecosystems, while weed flora is noticeably changing globally. In this study we evaluated the potential of cut-leaf ground-cherry (Physalis angulata L.), a species native to America, to invade the semi-arid regions of Greece. Greenhouse and laboratory experiments were conducted to evaluate the effects of different environmental resources (nutrient and water availability) on seedling growth, biomass production, fecundity, and seed germination of four populations of cut-leaf ground-cherry. Our results suggest that cut-leaf ground-cherry does not tolerate extreme drought during the first growth stages, while it can survive and produce adequate and rapidly germinated seed (> 85%) under low...

Involvement of a Host Erythrocyte Sialic Acid Content in Babesia bovis Infection OAK
TAKABATAKE, Noriyuki; OKAMURA, Masashi; YOKOYAMA, Naoaki; OKUBO, Kazuhiro; IKEHARA, Yuzuru; IGARASHI, Ikuo; 横山, 直明; 五十嵐, 郁男.
Host sialic acid (SA) has recently been suggested to play an important role in erythrocyte (RBC) infection by Babesia spp. The present study attempted to further determine the specific type of SAs important in the RBC invasion. Bovine RBC was found to bear abundant α2-3-linked SA residues but not α2-6-linked SA in nature, confirmed by flow cytometric analysis of the neuraminidase (Nm)-treated RBCs. Lectin-blot analyses revealed the removal of α2-3-linked SAs from the 97-, 33-, and 31-kDa bands by the Nm treatment. Addition of the Nm-treated RBCs into an in vitro culture of B. bovis resulted in a decreased population of the parasitized RBCs. The thin smear samples from the cultures were then observed under a confocal laser scanning microscope after staining...
